Gluten is the main group of proteins in grains such as wheat, rye, barley and to a lesser extent oats consisting of prolamins (in wheat: gliadin) and glutelins (in wheat: glutenins). Celiac disease is an immune-mediated enteropathy caused by the ingestion of gluten. A 33-mer peptide from alpha-2 gliadin has been identified as a principal contributor to gluten immunotoxicity. The shift in direction to detect immunotoxic peptides, that play a role in the pathogenesis of celiac disease, rather than simply the prolamins, led to the development of a next generation of antibodies. The G12 antibody was raised against this 33-mer and is a next generation antibody that targets a toxic fragment that triggers the auto-immune reaction in celiac patients.
