Gluten Analysis

Gluten is the main group of proteins in grains such as wheat, rye, barley and to a lesser extent oats consisting of prolamins (in wheat: gliadin) and glutelins (in wheat: glutenins). Celiac disease is an immune-mediated enteropathy caused by the ingestion of gluten. A 33-mer peptide from alpha-2 gliadin has been identified as a principal contributor to gluten immunotoxicity. The shift in direction to detect immunotoxic peptides, that play a role in the pathogenesis of celiac disease, rather than simply the prolamins, led to the development of a next generation of antibodies. The G12 antibody was raised against this 33-mer and is a next generation antibody that targets a toxic fragment that triggers the auto-immune reaction in celiac patients.

Gluten G12 AntibodyRomer Labs® has a complete portfolio for gluten testing. The monoclonal antibody G12 was used to develop a sandwich enzyme-linked immunosorbent assay (ELISA) called AgraQuant® Gluten G12 for quantitative testing in the laboratory and a lateral flow device (LFD) called AgraStrip® Gluten G12 for onsite factory testing, in addition to immunochemical test kits using polyclonal gluten antibodies. The G12 methods specifically measure the most immunotoxic proteins for those intolerant to gluten and thereby take analysis in a new direction in assuring food safety.
